    Two new tests rapidly identify Cryptococcus neoformans using phenoloxidase activity. These methods detect pigment production, enabling quick diagnosis of this yeast in clinical samples.

    Background:

    • Incorporation of caffeic acid into cornmeal agar for pigment detection.
    • Development of a non-medium-based test using L-beta-3,4-dihydroxyphenylalanine-ferric citrate paper strips.
    • Incubation of inoculated media/strips at 37°C for pigment observation.

    • Caffeic acid agar specifically produced brown pigment with C. neoformans isolates.
    • L-beta-3,4-dihydroxyphenylalanine-ferric citrate paper strips showed pigment production within 60-90 minutes for C. neoformans.
    • All other tested yeasts did not produce pigment in either test.

    • Both developed tests offer rapid and specific identification of Cryptococcus neoformans.

    • These tests can aid in the prompt clinical management of cryptococcosis.
