    Background:

    • Human fetal lung development is crucial for respiratory adaptation after birth.

    Purpose of the Study:

    • To investigate the presence and biochemical characteristics of a factor in human amniotic fluid that influences phosphatidylcholine synthesis.
    • To compare the properties of this amniotic fluid factor with known fibroblast-pneumonocyte factor.

    • Analysis of 41 human amniotic fluid samples (27-40 wk gestation).
    • Assay of [3H] choline incorporation into saturated phosphatidylcholine by human fetal alveolar Type II cells.
    • Biochemical characterization using Florisil stripping, acid-ethanol extraction, Sephadex G75 chromatography (acidic and neutral pH), and heat stability tests.

    • Activity decreased with advancing gestational age and was inversely related to cell number.
    • Chromatographic analysis revealed properties consistent with FPF, with higher activity in earlier gestations.

    • The biochemical properties of this factor are identical to fibroblast-pneumonocyte factor.
