Platelet markers in suicide attempters

Summary

Suicide attempters show reduced serotonin transporter (5HT) uptake and 3H-imipramine (3H-IMI) binding in platelets. These findings suggest a potential hypofunction in presynaptic serotonergic mechanisms linked to suicidal behavior.

Background:

Platelet 3H-imipramine (3H-IMI) binding sites and serotonin (5HT) uptake are analogous to brain mechanisms.
Sulphotransferase (ST) is an enzyme crucial for catecholamine metabolism.

Purpose of the Study:

To investigate alterations in platelet 3H-IMI binding, 5HT uptake, and ST activity in individuals who have attempted suicide.
To explore the potential link between these platelet markers and suicidal behavior.

Main Methods:

Measurement of 3H-IMI binding parameters in platelets.
Assessment of 5HT uptake kinetics in platelets.
Quantification of sulphotransferase (ST) activity in platelets.

A significant decrease in the density of 3H-IMI binding sites was observed in suicide attempters compared to controls.
Reduced 5HT uptake capacity was found in the platelets of suicide attempters.
No significant alterations in ST activity were detected between suicide attempters and healthy controls.

Conclusions:

The observed reduction in platelet 3H-IMI binding and 5HT uptake suggests a potential hypofunction of presynaptic serotonergic mechanisms in individuals with suicidal behavior.
These findings indicate that platelet markers may offer insights into the neurobiological underpinnings of suicidal behavior.
