[Macrophages in the choroid plexus of the embryonic telencephalon]
1Department of Morphology, Institute of Experimental Medicine, RAMS, St. Petersburg.
Abstract:
Using electron microscopy and immunocytochemistry distribution of macrophages in anlage of telencephalon choroid plexus was studied in 5-9 wks old human embryos. CD68 gene expression by intraepithelial macrophages that appear in anlage of choroid plexus (epithelial plate) was established to begin at 5th week of development. Accumulation of macrophages in embryonic connective tissue of choroid plexus anlage, appearance of intraepithelial macrophages, following appearance of free macrophages and supraependymal cells in brain ventricles (on wk 6-7) indicates that in human embryos choroid plexus anlage is a migration zone for macrophageal serie cells that penetrate into the cavity of the developing brain.
Insights
The choroid plexus is a key migration zone for macrophage cells in developing human brains, starting around the 5th week. These cells infiltrate the brain's cavities, contributing to early neural development.
Area of Science:
- Developmental biology
- Neuroscience
- Immunology
Context:
- The study investigates the early development of the choroid plexus in human embryos.
- Macrophage distribution and behavior within the developing brain are crucial for understanding neural development and disease.
- Previous research has not fully elucidated the role of the choroid plexus as a macrophage migration hub.
Purpose:
- To determine the origin and migration pathways of macrophages in the human telencephalon choroid plexus anlage.
- To establish the timeline of macrophage appearance and differentiation within the developing brain.
- To characterize the choroid plexus anlage as a potential site for immune cell entry into the fetal brain.
Summary:
- Electron microscopy and immunocytochemistry revealed macrophage distribution in the telencephalon choroid plexus anlage of human embryos (5-9 weeks gestation).
- CD68 gene expression confirmed the presence of intraepithelial macrophages in the choroid plexus anlage starting at the 5th week.
- Macrophage accumulation in connective tissue, followed by intraepithelial and free macrophages in ventricles (6-7 weeks), indicates the choroid plexus anlage is a critical migration zone for immune cells entering the brain.
Impact:
- This study identifies the choroid plexus anlage as a significant migration zone for macrophages in early human brain development.
- Findings provide insights into the early immune system's interaction with the developing central nervous system.
- Understanding these early immune cell dynamics may have implications for neurodevelopmental disorders and brain injury research.
