Sodium salt formation of mefenamic acid (MA) improved its formulation and dissolution properties. The resulting sodium mefenamate (Na-MA) tablets showed enhanced compressibility, hardness, and dissolution rates compared to MA alone.
Area of Science:
- Pharmaceutical Sciences
- Materials Science
Background:
- Mefenamic acid (MA) presents formulation and dissolution challenges.
- Developing improved dosage forms is crucial for therapeutic efficacy.
Purpose of the Study:
- To investigate sodium salt formation of mefenamic acid (Na-MA) to overcome its formulation and dissolution issues.
- To evaluate the physicochemical and performance characteristics of Na-MA compared to MA.
Main Methods:
- Sodium salt of mefenamic acid (Na-MA) was synthesized by reacting MA with sodium hydroxide.
- Salt formation was confirmed using Fourier Transform Infrared (FTIR) spectroscopy and Differential Scanning Calorimetry (DSC).
- Na-MA and MA were formulated into tablets with Avicel pH 101 and compressed; tablet properties (hardness, friability, capping, lamination) and dissolution rates were assessed.
Main Results:
- Na-MA powder formation was confirmed via FTIR and DSC.
- Tablets formulated with Na-MA exhibited superior direct compressibility, satisfactory hardness, and no capping or lamination.
- MA tablets showed capping and lamination issues, with friability failures.
- Na-MA tablets demonstrated significantly higher dissolution rates and extents across various media compared to MA tablets.
Conclusions:
- Sodium salt formation effectively addresses the formulation and dissolution problems associated with mefenamic acid.
- Na-MA offers a promising alternative for developing improved mefenamic acid dosage forms.
- The enhanced properties of Na-MA suggest improved bioavailability and therapeutic potential.
