Confocal Fluorescence Microscopy01:16

Confocal Fluorescence Microscopy

Confocal microscopy is an advanced microscopic technique. The prime advantage of the confocal microscope over other microscopy techniques is its ability to block the out-of-focus light from the illuminated samples using pinholes. It is widely used with fluorescence optics to obtain high-resolution, sharp contrast images. Unlike optical microscopes, confocal microscopes use a focused beam of light laser to scan the entire sample surface at different z-planes. These microscopes are, therefore,...

Summary

Laser capture microdissection (LCM) combined with immunocytochemical staining (immuno-LCM) enables precise cell isolation for molecular analysis. This powerful technique, when coupled with RT-PCR, allows sensitive gene expression studies from just 1-10 cells.

Area of Science:

  • Molecular Biology
  • Cell Biology
  • Genomics

Background:

  • Laser capture microdissection (LCM) isolates specific cells for molecular analysis.
  • Combining LCM with immunocytochemistry (immuno-LCM) enhances cell targeting.
  • Previous studies have applied these methods to various cell types and tissues.

Purpose of the Study:

  • To demonstrate the utility of immuno-LCM for isolating specific cell populations.
  • To evaluate the sensitivity of LCM combined with RT-PCR for gene expression analysis.
  • To showcase the application of these techniques in analyzing pituitary, thyroid, carcinoid tumor cells, and sarcomas.

Main Methods:

  • Immuno-LCM was used to label and isolate specific cells from dissociated tissues and cell cultures.
  • Reverse transcriptase polymerase chain reaction (RT-PCR) and Southern blot analysis were employed.
  • The methods were applied to heterogeneous cell populations including pituitary, thyroid, and carcinoid tumor cells, and sarcoma cells.

Main Results:

  • Immuno-LCM successfully isolated specific cell populations for molecular analysis.
  • The combination of LCM with RT-PCR enabled reproducible gene expression analysis from as few as 1 to 10 cells.
  • The study demonstrated successful application across diverse cell types and clinical samples.

Conclusions:

  • Immuno-LCM is a powerful tool for targeted cellular analysis.
  • LCM combined with RT-PCR significantly enhances the sensitivity of gene expression studies.
  • These integrated methods are valuable for cellular and molecular research, particularly in analyzing limited cell numbers.
