Glycogen synthase kinase 3 beta (GSK-3beta) gene variations may predict lithium treatment response in bipolar disorder patients. A specific GSK-3beta-50T/C polymorphism showed a higher T-allele frequency in lithium responders.
Area of Science:
- Neuroscience
- Genetics
- Psychiatry
Background:
- Glycogen synthase kinase 3 beta (GSK-3beta) is an enzyme targeted by mood stabilizers like lithium.
- Genetic variations (polymorphisms) in GSK-3beta may influence treatment efficacy.
Purpose of the Study:
- To investigate the association between GSK-3beta gene polymorphisms (-50T/C and -1727A/T) and lithium treatment response in Japanese bipolar disorder patients.
Main Methods:
- Genotyping of GSK-3beta-50T/C and -1727A/T single nucleotide polymorphisms (SNPs).
- Analysis of allele frequencies in lithium responders versus non-responders among 29 bipolar patients.
Main Results:
- A significant association was found between the GSK-3beta-50T/C polymorphism and lithium response.
- The T-allele of GSK-3beta-50T/C was significantly more frequent in lithium responders (P < 0.01).
- No significant association was observed for the GSK-3beta-1727A/T polymorphism.
Conclusions:
- The GSK-3beta-50T/C polymorphism may serve as a genetic marker for predicting lithium treatment response in bipolar disorder.
- Further research is warranted to validate these findings in larger cohorts.
