    Summary

This study tracked doublecortin (DCX) and NeuN protein expression in developing rat cerebellum neurons. Findings reveal distinct expression patterns crucial for assessing granular neuron development in early postnatal life.

    Area of Science:

    • Neuroscience
    • Developmental Biology
    • Immunohistochemistry

    Background:

    • Early postnatal development of the cerebellum is critical for motor function.
    • Understanding neuronal maturation requires specific molecular markers.
    • Doublecortin (DCX) and NeuN are key markers for neuronal development and maturation.

    Purpose of the Study:

    • To comparatively evaluate doublecortin (DCX) and NeuN expression in rat cerebellar cortex and nucleus interpositus neurons during early postnatal development (days 2-15).
    • To assess the utility of these markers in tracking granular neuron development.

    Main Methods:

    • Immunohistochemical analysis of DCX and NeuN expression in rat cerebellar tissue.
    • Comparative evaluation across different cerebellar regions (neocerebellum, paleocerebellum) and developmental time points (postnatal days 2, 7, 15).

    Main Results:

    • DCX expression was observed in migrating neurons of the cerebellar cortex, with higher levels in the neocerebellum at days 2 and 7.
    • NeuN expression appeared in migrating neurons and peaked in mature internal granular layer neurons.
    • Neither DCX nor NeuN were detected in Purkinje cells or the nucleus interpositus; NeuN expression increased in the nucleus interpositus over time.

    Conclusions:

    • The dynamic expression patterns of DCX and NeuN provide effective insights into granular neuron development in the early postnatal cerebellum.
    • This comparative immunohistochemical approach is valuable for assessing cerebellar neurodevelopmental processes.
