When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. Summary

Interleukin-25 (IL-25) is elevated in chronic rhinosinusitis with nasal polyps (CRSwNP), driving Th2 inflammation. Targeting IL-25 may offer a new therapeutic strategy for CRSwNP patients.

Purpose of the Study:

  • To investigate the association between IL-25 and Th2-biased inflammatory profiles in CRSwNP.
  • To evaluate IL-25 expression in nasal polyp tissues and its clinical relevance.

Main Methods:

  • Analyzed IL-25 expression in nasal polyp tissues from CRSwNP patients, chronic rhinosinusitis without nasal polyps (CRSsNP) patients, and controls using immunohistochemistry, immunofluorescence, flow cytometry, RT-qPCR, and ELISA.
  • Assessed inflammatory profiles and clinical characteristics of IL-25 high and low nasal polyp subtypes.
  • Examined the in vitro effects of IL-25 on Th2 cytokine production in cultured polyp cells.

  • Significantly increased IL-25 mRNA and protein levels were observed in nasal polyps compared to controls.
  • IL-25 high nasal polyps exhibited higher CT scan and endoscopic scores, alongside a pronounced Th2 response.
  • IL-25 exposure in vitro stimulated type 2 innate lymphoid cells and Th2 cells, enhancing Th2 cytokine production.

Conclusions:

  • Local IL-25 plays a critical role in promoting Th2-biased inflammation in nasal polyps.
  • IL-25 represents a potential therapeutic target for managing CRSwNP.
