    Background:

    • Suppressor T cells (Ts) play a critical role in immune regulation.
    • Antigen-presenting cells (APCs) are essential for initiating immune responses, including Ts cell induction.
    • The specific determinants on APCs that regulate Ts cell induction are not fully understood.

    Purpose of the Study:

    • To investigate the role of I-J and I-A determinants on splenic adherent cells (SACs) in the induction of suppressor T cells (Ts).
    • To determine the factors that maintain or restore the capacity of SACs to induce Ts cells after in vitro culture.
    • To elucidate the relationship between I-A and I-J expression on APCs and their function in Ts cell induction.

    Main Methods:

    • In vitro culture of antigen-coupled splenic adherent cells.
    • Assessing the ability of cultured cells to induce Ts cells.
    • Treatment with Concanavalin A (Con A)-stimulated lymphocyte supernatants and purified interferon-gamma (IFN-γ).
    • Analysis of I-A and I-J determinant expression using antibody-mediated elimination.
    • Flow cytometry to assess Ia expression.

    Main Results:

    • Antigen-coupled SACs lose the capacity to induce Ts cells after 4 days of in vitro culture.

    • Elimination of I-A-bearing SACs abrogates I-J-restricted Ts induction.

    Conclusions:

    • The induction of Ts cells by antigen-coupled SACs is critically dependent on I-J determinants.
    • In vitro culture leads to a loss of Ts-inducing capacity, associated with decreased I-A and I-J expression.
    • Interferon-gamma can restore Ts-inducing function by upregulating both I-A and I-J expression.
    • These findings suggest co-regulation of I-A and I-J on APCs, impacting the induction of Ts1 and Ts3 suppressor T cell subsets.
