Coptidis rhizome extract incubated with Rhodococcus sp. strain BD7100 yielded a new antibacterial compound, coptirhoquinone A. This compound shows antimicrobial activity against bacteria, fungi, and Pythium sp.
Area of Science:
- Natural Product Chemistry
- Microbiology
- Pharmacology
Background:
- Coptidis rhizome (CR) is a traditional herbal medicine containing protoberberine alkaloids.
- Rhodococcus sp. strain BD7100 degrades berberine (BBR), a component found in CR.
- CR extract incubated with strain BD7100 exhibited antibacterial activity.
Purpose of the Study:
- To isolate and identify novel compounds from CR extract incubated with strain BD7100.
- To elucidate the structure of the isolated compound.
- To evaluate the antimicrobial activity of the isolated compound.
Main Methods:
- Incubation of CR extract with Rhodococcus sp. strain BD7100 in Luria-Bertani (LB) medium.
- Isolation of coptirhoquinone A (1) using chromatographic techniques.
- Structure elucidation using Nuclear Magnetic Resonance (NMR) and Mass Spectrometry (MS).
- Total synthesis of coptirhoquinone A.
- Antimicrobial activity testing against bacteria, fungi, and Pythium sp.
Main Results:
- Coptirhoquinone A (1) was successfully isolated from the incubated CR extract.
- The structure of coptirhoquinone A was determined through NMR and MS analysis.
- Total synthesis of coptirhoquinone A was achieved.
- Compound 1 demonstrated antimicrobial activity against a range of microorganisms.
Conclusions:
- Coptirhoquinone A is a novel compound isolated from CR incubated with strain BD7100.
- The study provides a new avenue for discovering antibacterial agents from herbal medicines.
- Coptirhoquinone A exhibits broad-spectrum antimicrobial properties.
