Background:

  • Genomic DNA extraction is crucial for forensic analysis.
  • Forensic samples present unique challenges for DNA isolation.
  • The PrepFiler™ kits offer versatile solutions for diverse sample types.

Purpose of the Study:

  • To describe manual and automated DNA extraction methods using PrepFiler™ kits.
  • To detail the bind-wash-elute technique for genomic DNA purification.
  • To present protocols compatible with the AutoMate Express™ Forensic DNA Extraction System.

Main Methods:

  • Utilized PrepFiler™ Forensic DNA Extraction Kits (standard and BTA formats).
  • Employed a bind-wash-elute technique for DNA isolation.
  • Performed manual and semi-automated (AutoMate Express™) extraction protocols.

Main Results:

  • Optimized genomic DNA isolation and purification from various forensic samples.
  • Demonstrated effective DNA recovery from challenging sample types (bones, teeth, adhesives).
  • Purified DNA suitable for downstream forensic DNA analysis workflows.

Conclusions:

  • PrepFiler™ kits provide robust DNA extraction for forensic casework.
  • Both manual and robotic methods ensure efficient purification.
  • The AutoMate Express™ system streamlines the extraction process for forensic samples.
