Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

2.5K
Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl...

Genome-wide Association Studies-GWAS

12.3K
Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

Summary

Genetic variations in inflammation and endothelial function genes, specifically HABP2 rs7923349 and HABP2 rs932650, are linked to increased ischemic stroke risk. These gene interactions significantly elevate the likelihood of developing this condition.

Area of Science:

  • Genetics
  • Cardiovascular Research
  • Epidemiology

Background:

  • Ischemic stroke is a leading cause of death and disability worldwide.
  • Genetic factors play a role in stroke etiology, particularly those influencing inflammation and endothelial function.
  • Understanding these genetic correlations can aid in risk stratification and prevention strategies.

Purpose of the Study:

  • To investigate the association between genetic variations in inflammation and endothelial function-related genes and the risk of ischemic stroke.
  • To explore gene-gene interactions among these variants using generalized multifactor dimensionality reduction (GMDR).

Main Methods:

  • A multicenter, cross-sectional study involving 429 ischemic stroke patients and 429 matched controls from southwestern China.
  • Genotyping of 19 variants in 10 genes associated with inflammation and endothelial function.
  • Analysis of gene-gene interactions using GMDR.

Main Results:

  • Univariate analysis identified associations between stroke and variants rs7923349 (HABP2), rs8081248 (NOS2A), and rs932650 (HABP2).
  • GMDR analysis revealed significant gene-gene interactions between HABP2 rs7923349 and HABP2 rs932650.
  • High-risk interactive genotypes of HABP2 rs7923349 and HABP2 rs932650 were independently associated with a 3.578-fold increased risk of ischemic stroke (p < 0.001).

Conclusions:

  • Specific genetic variations in genes regulating inflammation and endothelial function are significantly associated with ischemic stroke.
  • The combined effect of high-risk genotypes in HABP2 rs7923349 and HABP2 rs932650 substantially elevates the risk of ischemic stroke.
