Sweet-responsive magnetic metal-organic framework for point-of-care testing of ATP using personal glucose meter

Abstract:
Personal glucose meter (PGM) is recognized as one of the most commercially available point-of-care testing (POCT) device for routine glucose monitoring. However, PGM-based analysis of non-glucose target is limited because of the complicated modification of glucose-related enzyme. Herein, we proposed a POCT sensing platform based on sweet-responsive magnetic MOF probe and PGM for the quantitative detection of non-glucose target. Glucose oxidase (GOx) and magnetic nanoparticles (MNPs) were co-encapsulated into the amorphous zeolitic imidazole framework-90 (aZIF-90) to form the probe and adenosine triphosphate (ATP) was chosen as the model target. The sensing probe couples the excellent affinity of aZIF-90 to ATP with the magnetic-responsive capability of MNPs, enabling the effective release and separation of free GOx. Under the assistance of PGM, the ATP level can be quantitatively determined by the GOx-based glucose consumption. A pretty wide detection range of 0.5-1000 μM and a low limit of detection of 81.01 nM were obtained. The sensing platform also displayed excellent detection performance in complicated real sample with recovery rates ranging from 93.98 % to 107.85 %. Therefore, this study provides an equipment-free and portable sensing platform for POCT sensing and sheds new light on repurposing PGM for non-glucose target analysis.
