Abstract:
Hu et al. identifies that Rho-associated coiled-coil containing kinase 2 (ROCK2) upregulation in sinusoidal endothelial and perivascular cells drives liver fibrosis via pro-fibrotic angiocrine signaling. The selective ROCK2 inhibitor TDI01 reverses fibrosis preclinically and demonstrates safety and target engagement in patients, highlighting vascular niche targeting as a therapeutic strategy.
