Owing to a lengthy career, I occupied a ringside seat to watch the evolution of our understanding of enzymatic catalysis. The field has progressed from primitive bioorganic models of enzyme active sites, which guided physical organic-based studies on how enzymes hydrolyze esters and amides, as well as laborious purifications of individual enzymes from animal organs, to investigations of multienzyme complexes, as featured in this text. My intent in this short essay is to provide background for the chapters in this volume. What papers and texts might have inspired the research reported within them? Where was the field at that time, and where might it be going?
