Linchpin reagents such as [1.1.1]propellane provide valuable opportunities to "Escape from Flatland" through the bioisosteric replacement of arenes with saturated hydrocarbons. Nevertheless, the direct incorporation of propellane-derived bioisosteres into peptides remains challenging. Herein we leverage photochemical conditions and the reactivity of [1.1.1]propellane to enable a versatile approach to cysteine-selective peptide modification.
