Developmental changes of mouse red cell pyruvate kinase.

    Summary

Mouse red blood cells show changes in pyruvate kinase (PK) activity and quantity during development. These findings suggest distinct PK forms exist in newborn versus adult mice.

    Area of Science:

    • Biochemistry
    • Developmental Biology
    • Hematology

    Background:

    • Red blood cell 2,3-diphosphoglycerate (2,3-DPG) levels increase during mouse postnatal development.
    • This increase correlates with a decrease in red cell pyruvate kinase (PK) activity.

    Purpose of the Study:

    • To investigate the changes in red cell pyruvate kinase (PK) during mouse postnatal development.
    • To determine if differences in PK activity are due to protein quantity or structural variations.

    Main Methods:

    • Immunoneutralization assays to quantify PK protein.
    • Cellulose acetate electrophoresis to analyze PK mobility.
    • Trypsinization to assess structural differences in PK.

    Main Results:

    • Newborn mouse red cells have higher PK activity, attributed to greater PK protein quantity.
    • Newborn and adult mouse red cell PK exhibit different electrophoretic mobilities.
    • Electrophoretic patterns vary between mouse strains (C57BL/6 and IS/Cam).
    • Trypsinization reveals structural differences in adult PK between strains.

    Conclusions:

    • Mouse red cells possess structurally distinct pyruvate kinase (PK) forms for newborn and adult stages.
    • These PK forms are analogous to human fetal gamma- and adult beta-globins.
