Background:

  • Diabetes mellitus is a complex metabolic disorder characterized by hyperglycemia.
  • Apoptosis, or programmed cell death, is increasingly recognized as a key factor in diabetes pathogenesis.
  • Understanding apoptosis mechanisms is crucial for improving therapeutic strategies in diabetic patients with comorbidities.

Purpose of the Study:

  • To investigate the mechanism of cell death initiation in rats with combined streptozotocin-induced diabetes mellitus and chronic enterocolitis.
  • To analyze the correlation between apoptotic cell markers and reactive oxygen species in this disease model.

Main Methods:

  • Experiments were conducted on 48 male rats divided into four groups: control, diabetes mellitus, chronic enterocolitis, and combined diabetes mellitus and chronic enterocolitis.
  • Apoptotic cells in blood leukocyte suspensions and reactive oxygen species were quantified using flow cytometry.
  • Correlation analysis was performed to assess relationships between various indices, with significance determined at p<0.05.

Main Results:

  • Combined diabetes and chronic enterocolitis significantly increased leukocytes with apoptotic signs compared to diabetes alone or enterocolitis alone.
  • The percentage of V+/PI- leukocytes increased by 24.2% versus the diabetes group and 88.0% versus the enterocolitis group.
  • V+/PI+ leukocyte values were 50.0% higher than in the chronic enterocolitis group, indicating enhanced cell death.

Conclusions:

  • The development of chronic enterocolitis combined with streptozotocin-induced diabetes in rats is associated with elevated leukocyte apoptosis.
  • Both free radical oxidation and inflammatory processes contribute to apoptotic death of leukocytes in this combined condition.
  • Reactive oxygen species demonstrate a stronger association with the development of apoptotic leukocyte death than inflammatory processes.
