Cleft lip and palate is the most common craniofacial anomaly. The etiology of orofacial clefts is multifactorial, encompassing a complex interplay of genetic and environmental influences, resulting in both syndromic and nonsyndromic clefts. The following discussion provides an overview of the most common syndromes implicated in cleft lip and palate, in addition to the genes and molecular pathways involved in craniofacial formation. Our goal is that continued understanding of these mechanisms will lead to standardized genetic screening and future advances in clinical transitional research.
